In a recent press release, pharmaceutical companies Merck and Moderna announced positive results from a late-stage trial of a novel skin cancer treatment that combines immunotherapy with mRNA vaccine technology. The treatment, known as intismeran, outperformed immunotherapy alone in reducing melanoma recurrence and limiting cancer spread in patients who had undergone prior melanoma surgery.
Although the Phase 3 trial is ongoing, preliminary results from an interim analysis indicated “statistically significant and clinically meaningful improvements,” according to the companies. Dr. Marcus Butler from Toronto’s Princess Margaret Cancer Centre expressed cautious optimism, highlighting the promising outcomes seen in the smaller Phase 2 study.
The treatment, which combines Merck’s immunotherapy drug Keytruda with a personalized mRNA vaccine from Moderna, targets abnormal proteins in cancer cells called neoantigens. This approach trains the immune system to identify and attack cancer cells bearing these antigens.
While the trial included over 1,100 high-risk patients with surgically removed Stage IIB-IV melanoma, concerns were raised about the potential immune-stimulating effects of mRNA vaccines beyond targeting melanoma neoantigens. The study’s focus on early-stage melanoma patients with completely removed tumors also raised questions about its effectiveness in later-stage cases.
As the trial progresses, further data will be presented at an oncology conference and shared with regulators. The success of this innovative treatment could have significant implications for melanoma patients, but more extensive studies and time are required to fully assess its efficacy across different disease stages.
